Simple method of evaluating scrapie in mice.

چکیده
Mice in advanced scrapie characteristically clasp their hindlegs together. As the disease progresses into the clasping stage, mice exhibit a sequence of identifiable hindleg reactions which have categorized and assigned relative scores permitting objective, quantitative evaluation of disease.

Scrapie agent proteins do not accumulate in grey tremor mice.
The grey tremor mouse is an autosomal recessive mutant characterized by a phenotype of unusual pigmentation, neurological abnormalities and early death. These mice have a spongiform encephalopathy similar to scrapie and Creutzfeldt-Jakob disease. Although the disease is clearly heritable, the grey tremor mouse spongiform pathology has also been transmitted by inoculation of genetically normal m...

Prions are unprecedented infectious pathogens that cause a group of invariably fatal neurodegenerative disease by an entirely novel mechanism. The conformational change in prion proteins results in a change from a predominantly α-helical protein to a β-sheet form, which causes scrapie in sheep and goat. The present study was carried out to identify polymorphisms of the prion protein gene (PrP) ...
متن کاملTubulofilaments in negatively stained scrapie-infected brains: relationship to scrapie-associated fibrils.
A simple method was devised for negative-stain transmission electron microscopy of brain infected with the agent of scrapie. Brains of infected hamsters contained large masses of tubulofilamentous structures with irregular fuzzy surfaces. Brains of mice infected with Creutzfeldt-Jakob disease agent contained similar tubulofilaments in smaller numbers. The abnormal tubulofilaments resembled but ...
متن کاملResistance of neonatal mice to scrapie is associated with inefficient infection of the immature spleen.
Previous studies demonstrated that neonatal mice up to about a week old are less susceptible than adult mice to infection by intraperitoneal inoculation with mouse-passaged scrapie. In peripherally inoculated adult mice, scrapie replicates in lymphoid tissues such as the spleen before invading the central nervous system. Here, we investigated scrapie susceptibility in neonatal mice in more deta...
متن کاملWestern blot detection of scrapie-associated fibril protein in tissues outside the central nervous system from preclinical scrapie-infected mice.
We describe a method of sample preparation to detect scrapie-associated fibril (SAF) proteins in small amounts of scrapie-infected mouse tissues by Western blot analysis using an antiserum to a synthetic peptide that corresponds to the N-terminal region of hamster prion protein. SAF proteins were efficiently detected in brain tissue by this procedure. The proteins were also detected in preparat...
